Fatty acid metabolism and hepatic lipogenesis.

چکیده
The conversion of octanoate-l-Cl4 to radioacetoacetate and the effect of pyruvate on this process are dependent on the K+ concentration in the incubation medium (1). These results prompted this investigation in which the incorporation of octanoate-l-Cl4 into fatty acids and cholesterol was determined in the presence of non-radioactive additions of pyruvate, succinate, and fumarate. Malonate was used as an aid in evaluating the relationship between the simultaneous metabolism of octanoate and the de novo synthesis of fatty acids. The lipogenic effects of high and low K+ concentrations in the medium were also studied.

Determination of Fatty Acid Oxidation and Lipogenesis in Mouse Primary Hepatocytes.
Lipid metabolism in liver is complex. In addition to importing and exporting lipid via lipoproteins, hepatocytes can oxidize lipid via fatty acid oxidation, or alternatively, synthesize new lipid via de novo lipogenesis. The net sum of these pathways is dictated by a number of factors, which in certain disease states leads to fatty liver disease. Excess hepatic lipid accumulation is associated ...
متن کاملEffect of randomly interesterified triacylglycerols containing medium- and long-chain fatty acids on energy expenditure and hepatic fatty acid metabolism in rats.
In our previous studies, medium- and long-chain triacylglycerols (MLCT), randomly interesterified triacylglycerols containing medium-chain and long-chain fatty acids in the same glycerol molecule, significantly reduced body fat accumulation in humans and rats. To clarify mechanism(s) for this effect of MLCT, we measured energy expenditure and hepatic fatty acid metabolism in rats by comparison ...
متن کاملAMP-activated protein kinase in the regulation of hepatic energy metabolism: from physiology to therapeutic perspectives.
As the liver is central in the maintenance of glucose homeostasis and energy storage, knowledge of the physiology as well as physiopathology of hepatic energy metabolism is a prerequisite to our understanding of whole-body metabolism. Hepatic fuel metabolism changes considerably depending on physiological circumstances (fed vs. fasted state). In consequence, hepatic carbohydrate, lipid and prot...
متن کاملThe Role of Akt2 in Hepatic Lipid Metabolism
Insulin drives the global anabolic response to nutrient ingestion, regulating both carbohydrate and lipid metabolism. When insulin resistance occurs in Type 2 Diabetes Mellitus, dysregulation of both of these processes ensues, resulting in hyperglycemia and lipid abnormalities. One of the most prevalent morbidities associated with insulin resistance is the abnormal accumulation of triglycerides...
متن کاملAltered Hepatic Lipid Metabolism Contributes to Nonalcoholic Fatty Liver Disease in Leptin-Deficient Ob/Ob Mice
Nonalcoholic fatty liver disease (NAFLD) is strongly linked to obesity, insulin resistance, and abnormal hepatic lipid metabolism; however, the precise regulation of these processes remains poorly understood. Here we examined genes and proteins involved in hepatic oxidation and lipogenesis in 14-week-old leptin-deficient Ob/Ob mice, a commonly studied model of obesity and hepatic steatosis. Obe...
متن کاملDietary olive oil and menhaden oil mitigate induction of lipogenesis in hyperinsulinemic corpulent JCR:LA-cp rats: microarray analysis of lipid-related gene expression.
In the corpulent James C. Russell corpulent (JCR:LA-cp) rat, hyperinsulinemia leads to induction of lipogenic enzymes via enhanced expression of sterol-regulatory-binding protein (SREBP)-1c. This results in increased hepatic lipid production and hypertriglyceridemia. Information regarding down-regulation of SREBP-1c and lipogenic enzymes by dietary fatty acids in this model is limited. We there...
